Nuclear war. We can shoot the missiles up before they blow up. Plague? Once its out, its pretty much stays out. Think a little like "Andromeda Strain" by Michael Crichton.

Not likely. Military power grows, but it can't hold a country captive to itself, and individual power grows too, maybe even faster than military power,  due to advancing technologies, though not to say that an individual's power is GREATER than the military's.

Actually I meant more along the lines of natural regulation; overpopulation, plague, famine et cetera.
And yes, if they are allowed to.

Natural regulation? Nope. Overpopulation is hardly a problem now other than individual specific areas, and we still have yet to expand into space. One possible technology ever so useful for colonizing Mars will be respirocytes and implanted CO2 scrubbers. Imagine being able to breathe the air of Mars, and hold your breath for HOURS?
In Short, with Transhumanism, we also transcend the flawed ideas of "nature" common in current society. The real truth of nature is that the entire universe, which includes all matter, energy, and etc. in it, is nature.

Possibly, if it is useful. I've never been much one for advancement for advancement's sake, so if there's no serious advantage posed by colonization, not particularly, no.

I think that happiness is a chemical reaction that can be achieved through a wide variety of means, including forcing the reaction to occur. So I suppose you could say that happiness comes only from this "oh-so-mortal" world
